We believe Indrepta A can benefit the health of all CF patients, including Class 1 CF mutations. Included in Indrepta A are powerful anti-inflammatories and antioxidants to reduce the inflammation and oxidative damage to tissue that fuels CF disease progression.
Class II CFTR mutations such as dF508del need help moving to the proper location within the cell. Indrepta ingredients (curcumin, resveratrol) included all three Indrepta versions are shown under experimental conditions to help with this movement. The dF508 allele also retains a very small amount of CFTR function. To benefit patients with any residual CFTR function, we include amentoflavone and forskolin in Indreptas B and C. These ingredients are well known in vitro to activate CFTR through non-corrector/potentiator mechanisms. Our research and reports by patients who carry residual function mutations that they can breathe better with Indrepta B over Indrepta A support the use of amentoflavone and forskolin for patients who carry residual function mutations.
Around 15% of CF patients carry one or more CFTR nonsense or ‘X’ mutations which cause CF disease through creation of a premature stop codon. Suppressing nonsense mutations by promoting read-through to produce full length functional CFTR is a therapeutic goal of Indrepta C. Curcumin, an ingredient in all versions of Indrepta, is also shown to improve function of the common nonsense mutation W1282X. 48
Escin and Boswellic acids were added into Indrepta C to achieve CFTR function by read-through from nonsense CFTR mutations (eg. W1282X, G542X and all others ending in X). However, Indrepta C is popular with other mutation classes who suffer from allergies, asthma or lung bleeds.
